Synthesis of 2-substituted pyrimido[5,4-b]indolizine derivatives from acetates of Baylis-Hillman adducts.

[摘要]：Novel tricyclic heterocycle pyrimido[5,4-b]indolizine derivs. I (R = Me, SMe, Ph, 3-MeOC6H4, 4-ClC6H4, 4-BrC6H4, 4-MeC6H4) were synthesized conveniently in two steps. The key intermediate 2-substituted 5-[(pyridin-2-yl)methyl]pyrimidin-4(3H)-ones, II, were obtained by facile cyclization of the acetate of Baylis-Hillman adduct III and various amidine hydrochlorides in the presence of sodium ethoxide. Treatment of II with phosphoryl chloride by reflux afforded I in moderate to high yields.
