Diversity-oriented synthesis of novel polycyclic scaffolds using polymer-bound reagents.

[摘要]A concise sequence utilizing a Petasis three component reaction followed by a tandem aza-Cope-Mannich cyclization afforded novel polycyclic heterocycles, e.g. I (R1 = H, Me; R2 = PhCH2, 4-MeOC6H4CH2, 5-methyl-2-thienylmethyl; R3 = Me, PhCH2, i-Bu, Me3COCH2, etc.; R4 = H, PhCH:CH, 2-benzofuryl; R5 = Me, Ph; R6 = H, Me), in good yields; alternative iminium cyclization based on a Pictet-Spengler reaction or aminal formation led to divergent pathways affording skeletal diversity.
