A photoreactive ruthenium(II) complex tethered to a guanine-containing oligonucleotide: A biomolecular tool that behaves as a "Seppuku Molecule".

[摘要]Self-destruction: Attachment of the photoreactive Ru(II) complex [Ru(tap)2phen]2+ (in which tap is 1,4,5,8-tetraazaphenanthrene and phen is phenanthrene) to a guanine-contg. oligonucleotide has led to an innovative biomol. tool for gene silencing studies. This conjugate can be selectively photo-cross-linked with its complementary strand but undergoes a self-inhibition (or "seppuku") process in the presence of a noncomplementary target, which is an attractive strategy to avoid undesired secondary photoeffects.
